On October 14, 2026, Verastem reported early results for VS-7375, its experimental oral KRAS G12D inhibitor, from the Phase 1/2 TARGET-D 101 trial. The data put it forward as a potential competitor in the race to develop KRAS-targeting pancreatic cancer treatments.
At the 900 mg daily dose, 4 of 8 patients with second-line pancreatic ductal adenocarcinoma had confirmed tumor responses (50%). Across 22 patients treated in the second-line-or-later setting, 9 had confirmed responses (41%), including one confirmed complete response and one unconfirmed complete response. The study is small and early-stage, so these figures need confirmation in larger trials.
Verastem said it had aligned with the FDA on a Phase 3 study design for first-line metastatic pancreatic cancer and expects to start the trial by year-end 2026. The planned study will compare VS-7375-based regimens with standard chemotherapy.
The news arrives as Revolution Medicines advances its own KRAS drug portfolio. Verastem’s results are promising, but the early data do not establish that VS-7375 is more effective than competing treatments.
Verastem also announced a $250 million financing and a CEO departure alongside the clinical update.
