Tango’s pancreatic cancer combo shows 82% response rate

At ESMO 2026, Tango Therapeutics reported updated Phase 1/2 results for vopimetostat plus daraxonrasib in previously treated pancreatic ductal adenocarcinoma with MTAP deletion and RAS mutation.

The combination produced an 82% objective response rate (18 of 22 patients). That is lower than the 92% reported in June among an earlier, smaller group, but the update included more patients; the results are still early and from a non-randomized study.

Median progression-free survival had not been reached, and 88% of patients were progression-free at nine months. Tango said all seven responses initially unconfirmed at the data cutoff were confirmed afterward.

Tango plans a registrational Phase 3 trial in front-line MTAP-deleted pancreatic cancer, aiming to start in the first half of 2027, subject to regulatory discussions. The planned trial is expected to compare the combination with standard chemotherapy.

Separately, vopimetostat plus zoldonrasib showed a 54% response rate in 35 evaluable patients with KRAS G12D-mutant pancreatic cancer. This is also early-stage evidence, not proof of benefit versus standard treatment.
