Newly disclosed FDA review data suggest Rasonque (daraxonrasib), Revolution Medicines’ pancreatic-cancer drug, may not work equally well across KRAS mutations. In the clinical-trial data, responses were strongest for tumors with KRAS G12V, more modest for the common G12D mutation, and weaker for G12R.
Because G12D is the most common KRAS mutation in pancreatic cancer, the variation raises questions about how broadly the overall trial results apply to patients with that subtype. The report says Revolution shares fell by more than 6% after the disclosure.
Analysts described the results as a possible opening for competitors developing mutation-specific G12D treatments or next-generation drugs. This is a potential competitive opportunity, not evidence that an alternative treatment has already proved superior.
The broader Phase 3 trial still showed a significant overall benefit versus chemotherapy:
median overall survival was 13.2 months with Rasonque versus 6.7 months with chemotherapy. The mutation-level findings add nuance to that overall result; they do not negate it.
