J&J touts promising early RIPTAC results in advanced prostate cancer

At ESMO 2026, Johnson & Johnson presented early Phase 1 results for JNJ-6143, an investigational oral drug using its RIPTAC platform, in heavily pretreated advanced prostate cancer.

Among 16 patients with measurable tumors, 12 (75%) had a confirmed complete or partial response. In the 50 mg dose group, 77% had a confirmed PSA drop of at least 50%, and 48% had a drop of at least 90%.

J&J oncology head Yusri Elsayed described the findings as the company’s best prostate-cancer drug data to date, calling them proof of concept for the RIPTAC approach.

The treatment appeared manageable in this early study:
reported common side effects included nausea and elevated cholesterol or triglycerides, and one patient discontinued because of an adverse event.

The results are promising but preliminary. The study was small, follow-up was short, and the reported figures do not establish how long responses last or whether the drug improves survival. J&J has moved forward with a Phase 2 study at 50 mg.
