At ESMO 2026, Incyte presented early Phase 1 results for INCB161734, its oral KRAS G12D inhibitor, combined with chemotherapy in metastatic pancreatic ductal adenocarcinoma (PDAC).
In the mFOLFIRINOX group, the company reported an 89% confirmed objective response rate, a 100% disease control rate, and a 90% six-month progression-free survival rate. The results were based on a small group of patients and are preliminary.
Incyte’s R&D head, Pablo Cagnoni, characterized the data as broadly comparable to Revolution Medicines’ zoldonrasib results, saying the companies were “neck and neck”—while acknowledging that cross-trial comparisons have limitations.
Incyte’s drug is an “ON/OFF” KRAS G12D inhibitor; Revolution’s zoldonrasib targets the “ON” state. Both companies are advancing combination treatment into late-stage trials.
Incyte said its Phase 3 study in first-line metastatic PDAC is underway, with topline results expected in 2028. The results are promising but do not establish that either treatment is superior.
