Pathos AI pads pipeline with $2.2B Alphamab deal and new AstraZeneca partnership

Pathos AI has signed a major deal with Alphamab Oncology worth up to $2.2 billion for the TROP2/HER3 bispecific antibody-drug conjugate JSKN016, including $125 million upfront and $2.09 billion in potential milestones. JSKN016 is in phase 3 for triple-negative breast cancer and other solid tumors. Pathos will develop and commercialize JSKN016 outside Greater China, while Alphamab retains rights within China, Hong Kong, Macau, and Taiwan.

Pathos AI has also announced a collaboration with AstraZeneca to advance AZD4241, an investigational oral PROTAC designed to degrade the estrogen receptor for ER+/HER2-negative breast cancer. Pathos will assume responsibility for early clinical development, expanding its pipeline to include three major oncology assets across four distinct tumor types.

Pathos’ pipeline now includes the PRMT5 inhibitor P-500 licensed from Prelude Therapeutics, the CBP/p300 inhibitor pocenbrodib from Novo Nordisk, and the MET-inhibitor DO-2 from its acquisition of DeuterOncology. All programs are identified or advanced via Pathos’ AI platform, Foundry, underscoring its AI-native drug-development strategy.

Alphamab will receive substantial non-dilutive funding from the upfront payment, with potential for milestone payments and royalties, while gaining a warrant to invest up to $62.5 million in Pathos. The deal is one of the largest licensing agreements for a Chinese biotech and highlights growing cross-border AI-driven oncology partnerships.

The JSKN016 and AZD4241 deals are expected to strengthen Pathos’ clinical oncology pipeline across breast, prostate, multiple myeloma, and MET-altered cancers, with both therapies planned for clinical development in the coming months.
