Fate Therapeutics is advancing an off-the-shelf iPSC-derived CAR T-cell program, FT819, for moderate-to-severe systemic lupus erythematosus and other B cell–mediated autoimmune diseases, with early data showing deep, sustained B-cell depletion and clinical signals of immune reset without intensive conditioning chemotherapy.
In a Phase 1/II study, FT819 has demonstrated no Grade ≥3 cytokine release syndrome (CRS), immune effector cell–associated neurotoxicity syndrome (ICANS), or graft-versus-host disease (GVHD), and can be dosed in an outpatient setting with same-day discharge, supporting a favorable safety profile in treatment-resistant autoimmune disease.
The company plans a potentially registration-enabling Phase II trial in lupus nephritis and is also developing next-generation off-the-shelf dual-CAR T candidates such as FT839 (targeting CD19 and CD38) tailored for multicellular autoimmune pathologies like rheumatoid arthritis and type 1 diabetes, underscoring early but promising clinical momentum for allogeneic CAR T therapy in refractory autoimmunity.
