Tango Therapeutics’ experimental PRMT5 inhibitor vopimetostat combined with Revolution Medicines’ RAS(ON) inhibitor daraxonrasib achieved a 92% objective response rate in an early‑phase trial of metastatic pancreatic ductal adenocarcinoma (PDAC).
The result comes from a Phase 1/2 dose‑escalation arm where patients received vopimetostat 200–250 mg once daily plus daraxonrasib 100 mg once daily.
In 12 response‑evaluable PDAC patients, 11 had an objective response (92%; 9 of 11 confirmed), 100% had disease control, and 90% were progression‑free at 6 months.
A second combination of vopimetostat plus another Revolution Medicines RAS(ON) inhibitor, zoldonrasib, achieved a 52% objective response rate in 27 patients with a 74% 6‑month PFS and 96% disease control.
The combinations were generally well tolerated:
no Grade 4–5 treatment‑related adverse events, no discontinuations due to toxicity, and most adverse events were Grade 1–2.
Analysts and company executives describe the PDAC vopimetostat–daraxonrasib data as unprecedented for this hard‑to‑treat setting and Tango plans to advance this combination into a Phase 3 first‑line trial for MTAP‑deleted pancreatic cancer later in 2026.
