City Therapeutics, a next‑generation RNA interference (RNAi) startup founded and led by former Alnylam executives, has raised $99.5 million in a Series B financing round to advance its first three RNAi programs and expand beyond traditional liver‑targeted applications.
Lead asset CITY‑FXI, an RNAi therapy targeting the blood‑clotting protein Factor XI, is already in Phase 1 clinical trials for thromboembolic diseases; the company expects to report its first Phase 1 data and initiate a Phase 2 trial later in 2026.
City is also advancing CITY‑RBP4 for Stargardt disease, a rare inherited retinal disorder, based on positive preclinical data; the company plans to file a clinical trial application and begin clinical testing by the end of 2026.
Part of the new capital will fund proprietary "cityRNA" trigger molecules and precision targeting ligands aimed at delivering RNAi to tissues beyond the liver, including the eye and central nervous system, building on recent collaborations with Biogen and Bausch + Lomb.
New investors Viking Global Investors and Sofinnova Investments joined existing backers ARCH Venture Partners, Regeneron Ventures, Rock Springs Capital, Fidelity Management & Research and others, bringing total funding for City Therapeutics to over $230 million since launch in 2024.
