A major theme is shifting from fresh, time-critical starting material to standardized cryopreserved leukapheresis, PBMCs, or purified T-cell inputs. This can decouple collection from manufacturing, improve plant scheduling, reduce site-to-site variability, and strengthen supply-chain resilience.
Recent evidence in CAR-T manufacturing indicates that cryopreserved PBMCs generally retain comparable expansion, transduction, and cytotoxic performance to fresh material, despite modestly lower post-thaw recovery/viability and occasional early delays in proliferation. A 2026 review reported post-thaw viability/recovery broadly in the ~77–97% range across studies.
JW Therapeutics reported Chinese approval to extend storage of sorted T cells used as starting material for Carteyva to 36 months. The company says the longer validated storage window supports inventory management, production planning, and more consistent manufacturing; its approach freezes purified CD4/CD8 T cells rather than whole PBMCs to reduce upstream variability.
Cryoport said its standardized, GMP-oriented IntegriCell cryopreservation service was selected by Verismo Therapeutics for clinical KIR-CAR-T programs. The announced goal is manufacture-ready cryopreserved leukapheresis material that can lower variability and help scale global operations. Cryoport also reported first shipments of cryopreserved clinical-trial material from its U.S. and Belgian facilities in Q1 2026.
Process technology is increasingly focused on closed, automated workflows:
controlled-rate freezing, automated fill/finish, standardized thawing, and in-process quality controls. These approaches aim to preserve cell yield and potency while cutting manual handling and enabling more reproducible batch performance.
New preservation chemistry is also a live development area. CaseBioscience launched a cGMP-aligned DMSO cryopreservation solution positioned around sustained post-thaw survival and functional performance. MIT researchers separately reported a sugar-based, lower-toxicity preservation approach for CAR-T-related cells that could reduce reliance on DMSO and simplify administration if validated further.
For off-the-shelf MSC and iPSC-derived therapies, recent technical discussions emphasize integrating cryopreservation with quality-by-design, potency assays, automation, and closed systems. The commercial objective is larger, more uniform batches that can be stored, distributed, and administered on demand.
